Electron Microscopy of Brain Tissue Fixed by Focused Beam Microwave to Preserve Glycogen

WhyGlycogen — the brain's main energy reserve — degrades rapidly post-mortem. Conventional chemical fixation fails to preserve its native levels, so this study uses focused beam microwave (FBMW) fixation as a rapid euthanasia + fixation method that better reflects in vivo conditions.
ConditionsPerfused = conventional PFA+GA perfusion (baseline). 4 kW · 600 ms and 5 kW · 600 ms = focused microwave irradiation settings applied at the head.
PipelineVolume EM stacks (Zeiss Volumescope) → manual + deep-learning segmentation (MitoNet, ResNet-Cardona, VGG19, DeepLab) → 3D reconstruction.
